Otokojuku, this manga focuses on Shishimaru Tsurugi, son of Momotaro Tsurugi from the previous series, and a new generation of Otokojuku students, while still featuring much of the same school staff. Years after the events of Ultimate Path Private High School 2011, Edajima Heihachi is informed of an invading alien race called the Susanoo, who seek to harvest the testicles of Japanese men to harness the Spirit of Yamato as an energy source. As a countermeasure, Edajima assembles all Otokojuku alumni, from the now-aging fighters from Sakigake!! Otokojuku, to the still young second generation from Akatsuki!! Otokojuku, and finally to the former Ultimate Path Private High School students who transferred to Otokojuku. Led by Tsurugi Momotaro, the Otokojuku alumni embark upon a quest to outer space to protect Japan from alien invaders.

Thirty years after World War III, Japan has suffered immense damage from a dangerous weapon known as a "Man Bomb" which affected nearly all Japanese men and rendered them unable to fight, with other men giving their lives for their country. Heihachi Edajima, an Otokojuku veteran and survivor of the Man Bomb, starts up a new facility known as Onnajuku, a tough combat training facility, and begins training several Japanese women to fight for their country. The manga follows Momoko Tsurugi, a descendant of former protagonist Momotarou Tsurugi, who recently enrolls in Onnajuku.
